Detector Function of Ultraviolet Spectroscopy

The UV detector measures the absorbance of ultraviolet light by a sample, converting it into a signal that quantifies the concentration of chromophoric compounds.Function of a UV Detector

In ultraviolet spectroscopy, the detector's primary function is to measure the intensity of UV light that passes through a sample and determine how much light is absorbed. When UV light interacts with a sample, electrons in chromophoric molecules are excited from a lower energy state to a higher energy state. The detector captures the resulting decrease in light intensity, which is proportional to the concentration of the absorbing species, allowing quantitative analysis of compounds in solution or eluent streams in HPLC systems .

Types of UV Detectors
  1. Fixed Wavelength Detectors: Measure absorbance at a single, preselected wavelength. They are simple and cost-effective but limited to compounds with known absorption maxima .
  2. Variable Wavelength Detectors (VWD): Use a deuterium lamp and a monochromator to select a specific wavelength in the UV range (typically 190–380 nm). The wavelength can be adjusted to match the absorption peak of the analyte, improving selectivity .
  3. Photodiode Array Detectors (PDA or DAD): Monitor the entire UV-Vis spectrum simultaneously using an array of photodiodes. This allows for quantitation, identification, and peak purity assessment of multiple compounds in a single run .
Working Principle

A UV detector typically consists of a light source (deuterium lamp for UV, tungsten lamp for visible), a monochromator to select the desired wavelength, and a photodetector to measure transmitted light. The detector compares the intensity of light before and after passing through the sample, producing an absorbance signal. This signal can be plotted against time or wavelength to generate spectra or chromatograms, which are used for qualitative and quantitative analysis .

Applications

UV detectors are widely used in HPLC, quality control, and analytical chemistry due to their reliability, ease of use, and universal response to chromophoric compounds. They are essential for determining concentrations, monitoring purity, and assessing stability of pharmaceuticals, biological macromolecules, and organic compounds .
